Overview of Phage Display

Phage display involves the presentation of antibody fragments on the surface of filamentous bacteriophages. Each phage particle carries the genetic information encoding its displayed antibody, creating a direct link between genotype and phenotype. Key phage display elements include:

  • Display format: scFv (single-chain variable fragment) or Fab
  • Phage system: Commonly M13; T7 or T4 may be used for larger inserts
  • Selection method: Biopanning using immobilized antigens, cells, or in vivo targets

6. Biopanning for High-Affinity Binders

We perform 3–5 rounds of panning under increasing stringency to isolate antigen-specific clones. Potential hits undergo phage ELISA screening before soluble expression.

7. Post-Selection Optimization

Validated sheep antibodies can be enhanced through:

  • Affinity maturation (mutagenesis + re-panning)
  • Humanization (grafting CDRs onto human frameworks)
  • Reformatting (IgG, Fc-fusions, bispecifics)
