Background: | Tumor necrosis factor alpha (TNF, TNFα, TNF-alpha) is a chemical messenger that acts as a mediator of the immune system and brings about inflammation. Primarily synthesized by activated macrophages, TNF triggers inflammation through its binding to the target receptors located on other cells. It is part of the tumor necrosis factor superfamily, a group of transmembrane proteins classified as immunocytokines—chemical messengers within the immune system. The overproduction of TNF is a key factor in various inflammatory disorders, including rheumatoid arthritis, psoriasis, and inflammatory bowel disease. In the treatment of these conditions, TNF-blocking drugs are commonly used; these medications work by stopping TNF from binding to its receptors. Additionally, TNF is linked to the pathological mechanisms of other diseases such as cancer, liver fibrosis, and Alzheimer’s disease, though the use of TNF inhibition has not yet yielded clear benefits in these cases. Because TNF plays an important and complex role in the immune system, its inhibition may give rise to side effects, such as an elevated risk of infections and the emergence of new types of autoimmunities. |
